ABSTRACT. We evaluated a method for the extraction and analysis, by Micellar Electrokinetic Capillary Chromatography (MECC), of all adenine nucleosides from human erythrocytes. The simultaneous detection of adenine nucleosides at different state of phosphorilation allows the calculation of the energy parameters ATP/ADP ratio and energy charge (EC). We have studied these parameters in the erythrocytes of cancer patients. The results obtained show an higher ATP/ADP ratio and an high energy charge in cancer patients with respect to normal controls. Although further experiments will be required to clarify these issues, the results reported here demonstrate the interesting potentiality of this analytic method, for a systematic study of energetic parameters in large populations such as neoplastic cachetic subjects and patients with AIDS.
Sulla possibile patogenicitą del virus trasmesso attraverso trasfusioni (TTV)

ABSTRACT. Among viruses assumed to be responsible of hepatitis in humans, transfusion - transmitted virus (TTV) is the last isolated in order of time (Japan, 1997). Its genome is characterized by a circular single strand DNA without envelope and with an unknown symmetry capside. Disagreeins are the hypothesis on its effective pathogenic role, both about a possible hepatic damage and its neoplastic evolution. Althoug TTV main transmission way is the parenteral one, its recovery in faeces, semen and saliva specimens give rise to a possible direct way of infection. There are no methods to reveal anti - TTV antibodies at the moment, and the only possible technique avaiable seems to be the Polymerase Chain Reaction (PCR).
a-Glutatione-s-transferasi del siero e danno epatico da alcool

ABSTRACT. The analytical performance of an enzyme immunossay for the measurement of serum a-glutathione-S-transferase (a-GST), and the predictive value of the test and its usefulness in the differential diagnosis between virus induced and alcool induced chronic active hepatitis, have been evaluated. Serum samples were obtained from 180 patients, including alcohol drinkers with liver disease, non drinkers patiens with liver disease, drinkers patients wihout liver disease, and healty subjects. The a-GST assay, whose reagents were obtained as a kit, was highly practicable, repeatible and reproducible; the test showed good specificity (95%) but low sensitivity (46-47%). Its a good marker for active liver damage, with significantly higher values in patiens with chronic active hepatitis in comparison to those with liver cyrrosis. It is also a good marker of viral chronic active hepatritis in comparison to alcohol induced hepatitis.
La preparazione dei vetrini in immunofluorescenza indiretta: valutazione di un sistema automatico

ABSTRACT. The indirect immunofluorescence (IFI) is a laboratory method for autoantibodies detection in use for several years. There are standardization problems in the reagent's nature, in the procedure and in the interpretation of the results. The aim of this study is a complete evaluation of an automated system to perform IFI method. We have compared HEP II slides performed in manual and by Hy Prep System Plus and we have verified no differences in ANA titers of examined sera. The precision of the instruments has been good and no carry over has been found . The microscopic cell fluorescence in slides automatically processed has been slightly worse, but it has not hampered the interpretation of cellular wells.
